Advanced IV Loading Dose Calculator

Estimate IV loading doses using core pharmacokinetic inputs. See calculations, tables, exports, and response curves. Designed for educational review before clinician verified medication decisions.

Educational use only: This calculator estimates a loading dose from entered pharmacokinetic values. It does not replace product labeling, local protocols, therapeutic drug monitoring, renal review, allergy checks, or clinician judgment.
IV assumption: Bioavailability is fixed at 1. Target concentration, Vd, and active fraction drive the estimate.

Calculator Inputs

Use the form below to estimate a generalized IV loading dose.

Example Data Table

Illustrative scenario only. This is not a medication order.

Scenario Target concentration Vd Weight Salt factor Stock strength Product dose Volume
Educational example 15 mg/L 0.7 L/kg 70 kg 1.00 50 mg/mL 735 mg 14.7 mL

Formula Used

This calculator estimates a generalized IV loading dose from a target plasma concentration and the total volume of distribution.

Active dose (mg) = Target concentration (mg/L) × Total Vd (L)

Product dose (mg) = Active dose ÷ Salt factor

If Vd is entered as L/kg: Total Vd (L) = Vd (L/kg) × Weight (kg)

Volume to administer (mL) = Product dose (mg) ÷ Stock strength (mg/mL)
For IV calculations, bioavailability is assumed to be 1. The salt factor adjusts the product amount when the formulation does not contain a full milligram of active drug per milligram of product.

How to Use This Calculator

  1. Enter the desired target concentration in mg/L.
  2. Choose whether the volume of distribution is entered as total liters or liters per kilogram.
  3. Add patient weight and choose kg or lb.
  4. Enter the salt factor. Use 1 if the product amount equals active drug amount.
  5. Add stock strength to estimate administration volume in mL.
  6. Set rounding increment and rounding mode if you want a practical rounded dose.
  7. Optionally enter a review threshold to flag large doses for manual review.
  8. Press the calculate button to display the result, summary table, and Plotly graph above the form.

Frequently Asked Questions

1) What does this calculator estimate?

It estimates a generalized IV loading dose from target concentration, volume of distribution, and active fraction. It is meant for educational review, not direct prescribing.

2) Why is bioavailability not editable?

This page is only for IV use. IV administration assumes complete systemic availability, so bioavailability is fixed at one.

3) When should I use L/kg for Vd?

Use L/kg when your source gives distribution volume relative to body weight. The calculator multiplies that value by weight to obtain total liters.

4) What is the salt factor?

It is the active-drug fraction of the product. A value below one means more product is needed to deliver the same active amount.

5) Why does the graph change with concentration?

The graph shows how the estimated rounded product dose and volume move as the target concentration changes across your chosen range.

6) Can this replace therapeutic drug monitoring?

No. Drug monitoring, renal assessment, formulation checks, infusion guidance, and clinical review remain essential for real patient care decisions.

7) Why is a rounded dose shown?

Rounded doses can be easier to prepare and document. The calculator still shows the raw estimate so you can compare both values.

8) What if I do not know the stock strength?

You can still calculate the dose in milligrams. The administration volume field remains blank until a stock concentration is entered.
