Protein purification is a series of processes intended to isolate one or a few specific proteins from a complex mixture, usually cells, tissues, or whole organisms. Quantitative analysis during these purification steps is critical for assessing the efficiency and success of the protocol. The foundational metric in this analysis is the total protein present within a given sample fraction.
During downstream processing, biochemists measure protein concentration using assays such as the Bradford assay, Lowry method, or UV spectrophotometry at 280 nm. However, concentration alone does not tell the whole story. To determine how much target protein has been recovered or lost across chromatography columns or precipitation steps, one must calculate the absolute mass of protein present in the entire volume of the fraction. This requires scaling the concentration by the total liquid volume and accounting for any sample dilution performed prior to the assay.
The calculation relies on a straightforward mathematical relationship combining concentration, volume, and dilution factors:
Total Protein (mg) = Concentration (mg/mL) × Volume (mL) × Dilution Factor
If your concentration is measured in micrograms per milliliter ($\mu g/mL$), the resulting total protein will be in micrograms, which can subsequently be converted to milligrams or grams depending on your analytical scale requirements.
